SUMMARY:

Triple negative breast cancer (TNBC) is commonly related for among 10% to 20% of all breast cancer diagnoses. CMTN have been characterized by the absence or low expression of estrogen (ER), progesterone (PR) and human epidermal growth factor type 2 (HER2 or ERBB2) receptors. The TNBC prognosis is unfavorable due to high metastasis rate, as well as the lack of targeted therapies. Therefore, the search for novel therapeutic strategies that are more selective and less harmful to patients need to be developed. One of the emerging promising alternatives consist on antimicrobial peptides (AMPs) utilization with antitumor potential. Such molecules can selectively act on the plasma membrane or on intracellular targets from neoplastic cells. Several studies have been carried out to elucidate the membranolytic mechanisms of action of these molecules, but only few studies have focused on gene expression modulation in neoplastic cells exposed to AMPs. Therefore, this work aims to evaluate MDA-MB-231 breast adenocarcinoma cell line gene expression alterations exposed to synthetic crotAMP14, which demonstrated in vitro antitumor activity. Furthermore, nuclear magnetic resonance spectroscopy studies have demonstrated that crotAMP14 presents an α-helix structural conformation at a membrane-mimetic environment. Relative gene expression analyzes by qRT-PCR suggest that crotAMP14 is capable of inducing the p53 expression, a central protein in the cell apoptosis process. Through cellular transcriptome analysis, data will be generated, allowing a better understanding of how metabolic pathways, genes or even non-coding RNA can be directly related to neoplasms development, shedding some light over AMPs molecular mechanisms of action.
