SUMMARY:

Depression can be attributed to an imbalance or deficiency of neurotransmitters, a hypothesis supported by the action of several classes of antidepressants, which block the reuptake of norepinephrine and/or serotonin and monoamine oxidase inhibitors (MAO). Brain-derived neurotrophic factor (BDNF) plays a crucial role in depression as a central regulator of neuroplasticity in the hippocampus. The drugs available for treatment have limitations, and studies have shown that ayahuasca (Aya) is a possible therapeutic option. Ayahuasca is an ancestral drink produced by cooking the Banisteriopis caapi vine, which contains the β-carbolines harmine, tetrahydroharmine (THH) and harmaline, MAO inhibitors, and the leaves of Pysicotria viridis, which contains N,N dimethyltryptamine (DMT), serotonin receptor agonist. Study previous study conducted by our group (Colaço et al., 2020) showed that the ayahuasca increased brain serotonin levels, the turnover of dopamine and BDNF in the hippocampus of Wistar rats. This project has aimed to investigate this potential using Wistar rats induced depression with lipopolysaccharide (LPS), a validated model for evaluation of new drugs through the quantification of monoamines by LC-MS/MS and BDNF in the hippocampus and evaluating behavioral parameters. Additionally, the number of neuronal progenitors will be evaluated, neurons and cells in cellular proliferation. Initially, 126 animals divided into 7 groups would be used: saline control, LPS control, Aya 0.5 (the dose. ritualistic) and LPS, Aya 1 and LPS, Aya 2 and LPS, positive control (fluoxetine and LPS) and DMT and LPS. After 14 days, the animals are subjected to open field and forced swimming tests, subsequently decapitated and the brain dissected to obtain the prefrontal cortex, nucleus accumbens, striatum and hippocampus, for analysis by LC-MS/MS for levels of dopamine, serotonin, norepinephrine and their metabolites, and immunofluorescence assays to quantify BDNF and cell proliferation markers. The blood collected is intended to assess the levels of inflammatory and anti-inflammatory cytokines.
