SUMMARY:

Several studies have demonstrated that in vitro maturation (IVM) induces a greater accumulation of lipids in bovine oocytes. This accumulation is not observed when maturation is performed in vivo. However, it is still unknown whether this difference in lipid content will remain after the oocytes are fertilized and cultured in vitro. Therefore, the objective of this study was/is to assess whether the maturation system affects embryo production and quality. For this, embryos produced in vitro from oocytes matured in vitro, in vivo (FSH and IFIOT) were used. The oocytes were matured in their respective maturation systems for twenty-two hours, and then were fertilized (IVF) and cultured (IVC) in vitro. The production rate and analyses of mitochondrial activity, lipid quantification, total number of cells and resistance to cryopreservation were compared among groups. Data related to blastocyst production rate, mitochondrial activity, lipid quantification and total cell number were analyzed by Chi-square and ANOVA (GLIMMIX) tests. For all analyzes a significance level of 5% (P<0.05%) was considered. As expected, the FSH group had a higher (P<0.05%) blastocyst rate than the IVM and TIFOI groups, which, in turn, were similar. Despite this, there was no difference between the total amount of cells and the mitochondrial activity of Bx in D7. However, the mean area occupied by lipids in IVM embryos (12.9%±7.73) is higher (P<0.05%) than in FSH (5.81%±4.36) and IFIOT (5.32%±4.08) embryos. Analyzes related to resistance to cryopreservation and the number of apoptotic cells have not yet been performed. At the present, the partial results demonstrate that IFIOT can be used as a tool for the maturation of oocytes in vivo, providing embryos similar to those produced in vivo and bringing new possibilities for reproductive technologies that require the in vitro maturation of oocytes.
