SUMMARY:

Cattle farming is one of the most important activities in the Brazilian agricultural sector. In 2022, the country continued to lead as the largest beef exporter in the world. This reality was only achieved by multiplication of the number of offspring from animals of greater genetic value, subsequent to the intensive use of reproductive biotechnologies. Brazil has become a reference in the development and commercial use of in vitro embryo production (IVEP) in cattle. Nevertheless, a number of quantitative and qualitative endpoints still need to be optimized. One example is the improvement of culture media, either by adjusting the physicalchemical properties, by adding agents with antioxidant potential, or by the supplementation with nutrients or sources of embryotrophic factors such as the fetal bovine serum (FBS). Supplementation with FBS has beneficial effects on in vitro embryonic development. However, its composition is undefined and may vary between batches. The use of FBS has been associated with metabolic alterations in embryos, with a potential negative impact on cryopreservation results. In addition, as it is a product of animal origin, there is an intrinsic risk of transmitting pathogens, as well as ethical issues related to its manufacturing process. Thus, it is necessary to search for alternatives to the use of SFB in embryo culture systems. This dissertation presents a review of protein supplementation during in vitro embryo culture, and the results of experiments aiming at use Yeast Extract (YE) as an alternative protein source to replace FBS during IVC. YE is known for improving cell culture systems of other mammals, as its composition contains important components for cell metabolism, in addition to presenting greater standardization among batches and negligible health risk, when compared to FBS. In general, the results obtained demonstrate the use of YE 1% (0.1mg/mL), as a supplement, doesn't compromise embryonic development or fresh post-transfer pregnancy rates, associated with BSA (3mg/mL) in sequential systems with low oxygen tension (5.5%), but YE causes greater accumulation of lipids in the cytoplasm.
