KEY WORDS:

Chagas disease; gut microbiota; Trypanosoma cruzi

PAGES: 35
BIG AREA: Ciências da Saúde
AREA: Farmácia
SUMMARY:

Chagas disease affects about 8 million people worldwide and is one of the main causes of death in Latin America. Considering the need to better understand aspects related to the pathogenesis of this disease for better therapeutic interventions and considering the growth of studies related to the impact of the intestinal microbiota in different pathologies, this study aims to evaluate the influence of T. cruzi infection on the intestinal microbiota of a murine model and to measure the impact of the use of antimicrobials on the course of CD. In this way, we use two different experimental models, Balb/c, susceptible to infection, and C57BL/6, resistant to infection, in acute and chronic phase, also separated into groups with intact microbiota and groups with modulated microbiota, infected with the Colombian strain. Regarding the applied methodology, qPCR was performed to analyze the composition of the intestinal microbiota for genera of bacteria (Lactobacillus sp., Enterococcus sp., Escherichia coli, Clostridium sp. and Bacteroides sp) and analysis of total bacteria, and parasitism in the heart and intestine, being also evaluated Th1 and Th2 profile cytokines by flow cytometry followed by histological analysis in order to identify the impact on the clinical evaluation of the mice in the heart and intestine tissue. The data obtained so far, indicate a possible impact of the intestinal microbiota in the process of regulation of T.cruzi infection with a role both in relation to cytokine production and modulation of different bacterial communities and the role of the genus Bacteroides in modulating the immune response in the infection resistant strain
