SUMMARY:

Chagas disease is an important public health problem in Latin America, where endemic areas coincide with the geographic distribution of triatomines. During the blood meal, the insect's bioactive salivary molecules ensure a continuous blood flow, exerting vasodilator, anti-inflammatory, antiplatelet, anticoagulant, immunomodulatory and anesthetic activity, which gives them great therapeutic potential. The identification of molecules from the saliva of these invertebrates revealed their diversity and abundance. The characterization of these molecules may contribute to the understanding of their roles along with other salivary components, as well as their potential as a target for vaccines aimed at hindering the blood meal. The objective of this project is to produce salivary proteins from triatomines in Leishmania tarentolae using the LEXSY system and perform the biochemical and functional characterization of the recombinant proteins. We are cloning the nucleotide sequences of the salivary proteins of interest into the plasmid pLEXSY_I-blecherry3 (Jena Bioscience), which will be used for L. tarentolae transfection. After selection of clones, proteins will be produced and purified by affinity chromatography. The purified proteins will be used to carry out tests to evaluate the inhibition of hemostasis.
