According to the Ministry of Agriculture, Livestock and Food Supply – MAPA, for the purpose of technical regulation, common bean, Phaseolus vulgaris and cowpea, Vigna unguiculata are considered as beans in Brazil, making these two cultures stand out in Brazilian agriculture. Despite the center of origin of cowpea being the western region of the African continent, Brazil has the fourth largest base collection of cowpea, around 4,000 preserved accessions. Embrapa Meio-Norte contains the largest active bank of cowpea in the country, including more than 3,500 accessions, in addition to cultivars that are recommended for small, medium and large growers. The cowpea genetic breeding program at Embrapa Meio-Norte has been working on the development of disease-resistant cultivars, with characteristics that allow mechanized harvesting and high grain productivity, increased resistance to pests and diseases, increased tolerance to high temperatures and water stress. Together with the technology created for the cowpea crop, phytosanitary problems were favored by the cultivation systems. Among these, diseases caused by plant-parasitic nematodes stand out. Cowpea can be attacked by several nematode species, especially nematodes of the genera Meloidogyne, Rotylenchulus and Pratylenchus. Among the root-knot nematodes (Meloidogyne spp.) M. incognita and M. javanica are the most important nematodes to the cowpea crop in Brazil. The use of resistant cultivars integrated 4 with appropriate crop rotation schemes has been considered one of the most sustainable methods for root-knot nematode management. However, root-knot nematode resistance has been controlled by the dominant Rk gene and the Rk2 gene, mapped to cowpea linkage group 9 with additive effect on the Rk gene, conferring a higher degree of resistance to virulent isolates. Even though, these two genes have been mapped, there is still a great demand for genes more effective for virulent nematode populations in cowpea crops. With the expansion of areas cultivated with cowpea and the occurrence of these nematodes in crop fields, it is essential to discover new genes that confer resistance to Meloidogyne spp., as well as the characterization of different nematode populations that could be used in the selection of resistance sources, mapping of resistance genes and study of the incompatible interaction between cowpea and Meloidogyne spp. In this sense, the objectives of this study are: to track and select cowpea accessions resistant to M. enterolobii and M. javanica; to map resistance genes; to study the transcriptome of bean plants showing resistance; and to study the incompatible plant-nematode interaction. Through mapping gene and studying the transcriptome, candidate genes for resistance can be found, or molecular markers associated with resistance that can be used in marker-assisted selection and even the insertion of these genes in other legume plants of economic importance, as soybean and beans. The results will make it possible to increase knowledge on the cowpea cultivars/ accessions developed by Embrapa Meio-Norte in relation to the response of these accessions to root-knot nematodes.
